چکیده

In this study, the suspension culture of Hortaea werneckii was incubated in supercritical carbon dioxide (SC CO2) in order to use the enzymes from these fungi for biotransformations and compare their activity with the activity of purified commercial enzymes at the same conditions. H. werneckii cell suspension was treated in SC CO2 at 35 °C and different pressures. The survival of H. werneckii cells was determined by measuring the optical density of the cell suspension. Additionally, the total protein concentration and absorbance of nucleic acids in the suspension were also determined. The interest was to excrete the intracellular enzyme α-amylase from H. werneckii and to keep the extracellular enzyme cellulase in the active form after the treatment of the cell suspension culture with SC CO2. The activity of intracellular enzyme eliminated from H. werneckii cells and that of extracellular enzyme after the treatment of the cells with SC CO2 was compared with the activity of commercial available enzymes. A high amount of proteins was released from H. werneckii cells during the SC CO2 treatment. The residual activity of both examined enzymes decreased with prolonged treatment time of H. werneckii cell suspension in SC CO2, but both could be still used as biocatalysts in this medium.

Production and Characterization of Melanin Pigment from Halophilic Black Yeast Hortaea werneckii

Melanin is nearly a ubiquitous pigment synthesized by living organisms in the course of hydroxylation and polymerization of organic compounds. Melanins have immense application potentials in the field of agriculture, cosmetics and pharmaceutical industries (photoprotection and mosquitocidal activity isolated from Streptomycete). In this study, the halophilic black yeast, Hortaea werneckii produ...

The unique characteristics of HOG pathway MAPKs in the extremely halotolerant Hortaea werneckii.

HwHog1A/B, Hortaea werneckii homologues of the MAP kinase Hog1 from Saccharomyces cerevisiae, are vital for the extreme halotolerance of H. werneckii. In mesophilic S. cerevisiae, Hog1 is phosphorylated already at low osmolyte concentrations, and regulates expression of a similar set of genes independent of osmolyte type. To understand how HwHog1 kinases activity is regulated in H. werneckii, w...

Hortaea werneckii isolated from silicone scuba diving equipment in Spain.

During a survey of black yeasts of marine origin, some isolates of Hortaea werneckii were recovered from scuba diving equipment, such as silicone masks and snorkel mouthpieces, which had been kept under poor storage conditions. These yeasts were unambiguously identified by phenotypic and genotypic methods. Phylogenetic analysis of both the D1/D2 regions of 26S rRNA gene and ITS-5.8S rRNA gene s...

Preparation of PMMA/MWNTs Nanocomposite Microcellular Foams by In-situ Generation of Supercritical Carbon Dioxide

Nanocomposites containing poly(methyl methacrylate) (PMMA) and surface functionalized Multi-Walled Carbon Nanotubes (MWNTs) were synthesized. The dispersion of MWNTs in PMMA was characterized using Transmission Electron Microscopy (TEM).The synthesized nanocomposites were successfully foamed using a simple method based on the in-situ generation of supercritical carbon dioxide (CO2</sub...
